Vigabatrin


General
  Clinical Use: Vigabatrin (Sabril) is an anticonvulsant, GABA (Gamma-Aminobutyric Acid) Transaminase inhibitor, used for adjunctive therapy in epilepsy not controlled by other epileptic drugs. Vigabatrin can cause progressive and permanent visual field defects in a high percentage of patients and should not be used concomitantly with retinotoxic drugs. Other adverse events are mainly CNS-related, such as sedation, somnolence, fatigue and impaired concentration.

Vigabatrin plasma concentrations are related to dosage, but there is no strong correlation between plasma concentration and clinical effects. Out of 47 vigabatrin requests - 64% of these samples had plasma levels between 0 and 100 umol/L, 30% were between 100 and 200 umol/L, and 6% were between 200 and 700 umol/L. Trough levels were found within the range 19-43 umol/L (with a mean of 34 umol/L)
