Amelogenin Sex Typing PCR

Clinical Use:
|
This PCR is used to monitor the engraftment status of a patient after BMT.
It uses the amelogenin gene, which has an X and Y homologue that differs by a 6 base-pair deletion in the X homologue, to determine if male or female DNA is present by presence or absence of the Y homologue. This is used to indicate the presence of donor and/or recipient DNA, thereby indicating the source of reconstitution of the bone marrow after transplant.
Samples of donor and recipient are required prior to transplant to confirm the expected sex amelogenin banding pattern for each source of DNA. Peripheral blood samples are then collected from the patient post transplant to monitor the engraftment status. The progressive change in DNA from recipient to donor (indicated by the change in the banding pattern of the PCR products) is indicative of successful marrow engraftment.
Persistent or reappearance of recipient DNA is suggestive of failure to engraft with donor cells or relapse. Persistence of both donor and recipient DNA indicates a mixed chimaeric state.
This PCR is only performed if PCRs for VNTR is not informative. |

A sample from the transplant recipient AND donor must be received before transplant.
If recipient DNA has not been stored before BMT, a 10mL saline mouthwash to provide buccal cells must be obtained post BMT as a source of recipient DNA. |
